Southgate has left out Arsenal forward Eddie Nketiah and AC Milan defender Fikayo Tomori from his squad to face Italy at Wembley Stadium, with 10 changes expected.
He had to trim his squad as nations are only allowed 23 players on matchdays.
That's according to Rob Dorsett, who tweeted: "#afc Eddie Nketiah and #milan Fikayo Tomori have been left out of the England squad for tonight's Euro qualifier against Italy.
"Southgate had 25 players for the final training session at Spurs training ground yesterday, but nations are only allowed to name a matchday squad of 23."
Nketiah made his England debut against Australia last Friday while Tomori started the friendly before being replaced in the second-half.
This season, Nketiah has scored two goals in eight league appearances for the Gunners.
As for Tomori, he's played seven Serie A games this campaign.
The Three Lions, meanwhile, beat Australia 1-0 thanks to a second-half strike from Aston Villa's Ollie Watkins.
They take on Italy as they look to secure their spot for next summer's European Championship in Germany.
England have won four of their five Euro qualifiers and sit at the top of Group C, with 13 points on the board, three points ahead of Italy.
