Gyokeres scored the second goal in their 2-0 win against Rio Ave at the Jose Alvalade Stadium after dinking a penalty down the middle.
The striker has scored 20 consecutive spot-kicks for the club since his one and only miss last February.
Sporting CP will thus take a two-goal advantage into the second leg later this month.
It is all set up for a Lisbon derby in the final.
In the other semi-final, Benfica plays fourth-tier Tirsense, the lowest-ranked side ever to reach this stage of the cup.
Sporting CP lost the final to Porto after extra time last year in a rematch of 2019, when the former won following a penalty shootout.
One of the Lisbon rivals will also win the title, with just goal difference separating them.
Gyokeres will hope to help Sporting CP retain their crown and do the double after already coming up short in the League Cup final and Super Cup thus far this season.
He has them already halfway to another final, with his head coach Rui Borges acknowledging after the semi-final first leg:

"He has helped the team to grow, and he gives confidence to his teammates. He trains like the rest.
"Above all, he is a focused player who pays attention to the details; he's competitive, never lets his guard down and doesn't want to rest. Maybe he could have rested today.
"He is a different player; he is clearly the best in our league. It is a young team, and as an athlete, player and teammate, he inspires confidence in his teammates with what he brings to the team."
Those words will be music to the ears of Arsenal fans. The club's new sporting director, Andrea Berta, has reportedly prioritised signing the Sweden international during the summer transfer window.
